Both are first-line blood pressure medicines and they act on completely different systems. Amlodipine is a calcium channel blocker: it stops calcium entering the smooth muscle of artery walls, so the vessels relax directly. Lisinopril is an ACE inhibitor and works hormonally, reducing the production of angiotensin II.
